I'm surprised Adobe Illustrator on his list. That would have been ideal for this project.
I think he may have used a tablet too. He may have used a tablet for the initial sketch. Everything else he did there can be done with a mouse.

EDIT: if he was using a tablet, he could've overlayed an image he already drew and been tracing it. It'* also been sped up.
