Does anyone know if GM makes the SES light come on ..on a 2000 *-10 ZR2 at 50k miles?
I was thinking it may be for transfer case/ tranny fluids.
I have access to a OBD II scan tool....will that tell me ...and can I shut that light off with it..

The scan tool will tell you the code, and also give a short description of the problem. For a better description of the problem you need the GM service manual.

The most common cause of a 4.3 (assuming it had the W or Z engine) SES light is a clogged EGR. This can be removed and cleaned with brake cleaner but you will need to have a new gasket. Check the codes first. These things have a lot of computer controlled electronics that can through a SES.
