If I upgrade my 2016 Buick enclave standard radio with a navigation radio that I bought do I have to carry it to the dealer to get it repaired or flashed for my vehicle?

Yes, the VIN will need to be programmed in. If you swap it now before leaving, you may not be able to start your car.
